Can't connect through LAN

I run a network of 30 client machines, connecting to the internet through our
LAN server. However, one machine has suddenly stopped being able to connect.
All other network functions are still fine, as is email, which points to
Internet Explorer being the problem. I've run the Internet Connection Wizard
again (several times in fact) to reset the LAN connection via a proxy server
to no avail.
What am I missing?
