Don,
Did you check the control panel entries in safe mode? How about the control
panel /sounds, video and game controllers--did you check there? Is there a
wire running from the mobo to the sound jack on the back of the case? If
so, did the wire get knocked off when you replaced the floppy drive? Any
chance you can try the speakers on another PC?

Aargh . . .
That did it: speakers work fine on the old PC upstairs, but are
dead on the newer system: must be the SOB component
(the SOB! I did not expect this from ASUS hardware.)
