I'm using a Microsoft  Wireless Optical Mouse with a Windows ME laptop. This
device is connected to a Targus USB-hub together with a webcam and a
MP3-player. The problem is that sometimes (usually after the computer have
been left on for some time) the light on the mouse transmitter shutdowns and
the mouse stop responding. The computers built in touchpad still works and so
are the other devices on the USB hub.  When you restart the computer
everything works fine again but how can I get it to work again without
restarting the system?

Make sure the USB hub is independently powered.  If not, obtain same.
